Abstract: Fluorescence °uctuation spectroscopy is widely used in modern life sciences. Study of molecular structure and dynamics in the systems like cell membranes with di®usion limited in two dimensions needs a modiЇcation of the existing analysis methods. Here we present a modiЇcation of the photon counting his- togram method, most commonly used for the intensity distribution analysis in °uorescence °uctuation spectroscopy, for the case of a two-dimensional brightness proЇle.
